Nile Blue Staining of Cells in Amniotic Fluid for Fetal Maturity

Abstract

Many methods for evaluating fetal maturity have been tested, and, although the staining of cells in amniotic fluid with Nile blue dye has been advocated by several investigators, severe criticism has been afforded this method of fetal maturity assessment. In this study the six commercially available brand!) of Nile blue dye were used to estimate the length of gestation. These dyes differed in the type of dye, color index, solvent base, and percentage of Nile blue present. Only two of the six stains gave acceptable results in determinig fetal maturity in some 40 samples of amniotic fluid. It would appear that the choice of dye may explain the paradoxical results presented by other studies. In addition, many factors germane to this technic are discussed.
