Effects of Medroxyprogesterone Acetate on Carbohydrate Metabolism

The carbohydrate metabolism of 18 women receiving long-term treatment with medroxyprogesterone acetate (MPA) and 14 control women was investigated. Only 1 subject in the treated group had an impaired glucose tolerance and this was accompanied by excessive weight gain. The MPA caused an increase in scrum insulin which was significant at the fasting and 3-hour periods. Serum growth hormone levels tended to increase, but the difference was significant at the 3-hour period only. The drug had no clear effect on plasma free fatly acids or serum triglycerides. In view of the hyperinsulinism, it is concluded that MPA is diabetogenic and the implications of this are discussed.
